YM 155 is the first-in-class surviving inhibitor with an IC50 0.54 nM, selectively inhibits survivin expression and induces apoptosis in tumor cells. YM 155 is in clinical trials. Survivin is a member of the inhibitor of apoptosis (IAP) gene family, and is expressed during embryonal development, but is absent in most normal, terminally differentiated tissues. Survivin is upregulated in a variety of human cancers. Expression of suvivin in tumors is associated with a more aggressive phenotype, shorter survivial times, and a decreased response to chemotherapy.[1][2][3]
