AST-6, a rationally designed novelanilinoquinazoline compound, inhibited enzymatic activitiesof wild type EGFR and ErbB2 as well as EGFR L858R/T790M resistantmutant both in cell-free and cell-based levels. Importantly, AST6 functions as an irreversible inhibitor, most likely bycovalent modification of Cys 797 and Cys 805 in the catalyticdomains of EGFR and ErbB2, respectively. Further studies showedthat AST 6 inactivated their downstream pathways and therebyinhibited the proliferation of a panel of cancer cell lines. [1]
