Flavopiridol (Alvocidib, HMR-1275, L-868275) is a pan-CDK inhibitor targeting CDK1, CDK2, CDK4, CDK5, CDK6, and CDK9 with IC50 of 30 nM, 170 nM, 100 nM, 170 nM, 80 nM and 20 nM, respectively. It inhibits the phosphorylation of cyclin-dependent kinases (CDKs) and suppresses expression of cyclin D1 and D3, interrupting the central role of CDKs in the regulation of the cell cycle and consequently promoting apoptosis. This action has been exploited in studies investigating the suppression of oncogenic proliferation, synovial hyperplasia, and joint destruction related to rheumatoid arthritis. [1][2]
