Pemetrexed disodium (Alimta, LY-231514) is a novel antifolate and antimetabolite for TS, DHFR, and GARFT with Ki of 1.3 nM, 7.2 nM and 65 nM, respectively. Pemetrexed binds to and inhibits the enzyme thymidylate synthase (TS) which catalyses the methylation of 2'-deoxyuridine-5'-monophosphate (dUMP) to 2'-deoxythymidine-5'-monophosphate (dTMP), an essential precursor in DNA synthesis. ^[1][2]
